    Contributions on agreement in dynamic distributed systems

    139 p.This Ph.D. thesis studies the agreement problem in dynamic distributed systems by integrating both the classical fault-tolerance perspective and the more recent formalism based on evolving graphs. First, we developed a common framework that allows to analyze and compare models of dynamic distributed systems for eventual leader election. The framework extends a previous proposal by Baldoni et al. by including new dimensions and levels of dynamicity. Also, we extend the Time-Varying Graph (TVG) formalism by introducing the necessary timeliness assumptions and the minimal conditions to solve agreement problems. We provide a hierarchy of time-bounded, TVG-based, connectivity classes with increasingly stronger assumptions and specify an implementation of Terminating Reliable Broadcast for each class. Then we define an Omega failure detector, W, for the eventual leader election in dynamic distributed systems, together with a system model, , which is compatible with the timebounded TVG classes. We implement an algorithm that satisfy the properties of W in M. According to our common framework, M results to be weaker than the previous proposed dynamic distributed system models for eventual leader election. Additionally we use simulations to illustrate this fact and show that our leader election algorithm tolerates more general (i.e., dynamic) behaviors, and hence it is of application in a wider range of practical scenarios at the cost of a moderate overhead on stabilization times

    Routing in MobileWireless Sensor Networks: A Leader-Based Approach

    This paper presents a leader-based approach to routing in Mobile Wireless Sensor Networks (MWSN). Using local information from neighbour nodes, a leader election mechanism maintains a spanning tree in order to provide the necessary adaptations for efficient routing upon the connectivity changes resulting from the mobility of sensors or sink nodes. We present two protocols following the leader election approach, which have been implemented using Castalia and OMNeT++.     Treatment with tocilizumab or corticosteroids for COVID-19 patients with hyperinflammatory state: a multicentre cohort study (SAM-COVID-19)

    Objectives: The objective of this study was to estimate the association between tocilizumab or corticosteroids and the risk of intubation or death in patients with coronavirus disease 19 (COVID-19) with a hyperinflammatory state according to clinical and laboratory parameters. Methods: A cohort study was performed in 60 Spanish hospitals including 778 patients with COVID-19 and clinical and laboratory data indicative of a hyperinflammatory state. Treatment was mainly with tocilizumab, an intermediate-high dose of corticosteroids (IHDC), a pulse dose of corticosteroids (PDC), combination therapy, or no treatment. Primary outcome was intubation or death; follow-up was 21 days. Propensity score-adjusted estimations using Cox regression (logistic regression if needed) were calculated. Propensity scores were used as confounders, matching variables and for the inverse probability of treatment weights (IPTWs). Results: In all, 88, 117, 78 and 151 patients treated with tocilizumab, IHDC, PDC, and combination therapy, respectively, were compared with 344 untreated patients. The primary endpoint occurred in 10 (11.4%), 27 (23.1%), 12 (15.4%), 40 (25.6%) and 69 (21.1%), respectively. The IPTW-based hazard ratios (odds ratio for combination therapy) for the primary endpoint were 0.32 (95%CI 0.22-0.47; p < 0.001) for tocilizumab, 0.82 (0.71-1.30; p 0.82) for IHDC, 0.61 (0.43-0.86; p 0.006) for PDC, and 1.17 (0.86-1.58; p 0.30) for combination therapy. Other applications of the propensity score provided similar results, but were not significant for PDC. Tocilizumab was also associated with lower hazard of death alone in IPTW analysis (0.07; 0.02-0.17; p < 0.001). Conclusions: Tocilizumab might be useful in COVID-19 patients with a hyperinflammatory state and should be prioritized for randomized trials in this situatio

    The endocannabinoid system and PPARγ are important targets for the development of novel compounds against fibrotic diseases such as systemic sclerosis (SSc), also called scleroderma. The aim of this study was to characterize VCE-004.3, a novel cannabidiol derivative, and study its anti-inflammatory and anti-fibrotic activities. The binding of VCE-004.3 to CB1 and CB2 receptors and PPARγ and its effect on their functional activities were studied in vitro and in silico. Anti-fibrotic effects of VCE-004.3 were investigated in NIH-3T3 fibroblasts and human dermal fibroblasts. To assess its anti-inflammatory and anti-fibrotic efficacy in vivo, we used two complementary models of bleomycin-induced fibrosis. Its effect on ERK1/2 phosphorylation induced by IgG from SSc patients and PDGF was also investigated. VCE-004.3 bound to and activated PPARγ and CB2 receptors and antagonized CB1 receptors. VCE-004.3 bound to an alternative site at the PPARγ ligand binding pocket. VCE-004.3 inhibited collagen gene transcription and synthesis and prevented TGFβ-induced fibroblast migration and differentiation to myofibroblasts. It prevented skin fibrosis, myofibroblast differentiation and ERK1/2 phosphorylation in bleomycin-induced skin fibrosis. Furthermore, it reduced mast cell degranulation, macrophage activation, T-lymphocyte infiltration, and the expression of inflammatory and profibrotic factors. Topical application of VCE-004.3 also alleviated skin fibrosis. Finally, VCE-004.3 inhibited PDGF-BB- and SSc IgG-induced ERK1/2 activation in fibroblasts. VCE-004.3 is a novel semisynthetic cannabidiol derivative that behaves as a dual PPARγ/CB2 agonist and CB1 receptor modulator that could be considered for the development of novel therapies against different forms of scleroderma

    Resumen basado en el de la publicación.A lo largo del curso 2012/2013 se ha desarrollado en el IES Eduardo Valencia de Calzada de Calatrava el Proyecto de Innovación Educativa “Patrimonio local y turismo rural: Calzada de Calatrava y Aldea del Rey”, con el fin de que los alumnos profundizasen en el conocimiento de los recursos histórico-artísticos, naturales y antropológicos que ambas localidades ofrecen, pusieran en valor el patrimonio que posee el medio que les rodea, y apreciasen sus potencialidades económicas y laborales, que pueden resultar básicas para la dinamización de la comarca. También se perseguía mostrar la importancia del aprovechamiento de las posibilidades que el conocimiento de idiomas extranjeros, el dominio de las tecnologías de la información y el uso del diseño otorgan a los alumnos, que han de constituirse en futuros recursos laborales.
